Practical advice:
iPS reprogramming can be optimized by varying the amount of Klf4 to achieve maximal reprogramming efficiency. Generally, the multiplicity of infection (MOI) of KOS and c-Myc vectors should be kept at a 1:1 ratio, while the Klf4 vector can be varied independently. The standard MOI ratio for KOS:c-Myc:Klf4 is 5:5:3. To improve reprogramming efficiency, this can be changed to 5:5:6 or 10:10:6.
